Cu2+ 、Zn2+ 固载硅胶整体柱对8-羟基-2'-脱氧鸟苷固相微萃取性能研究
摘要:
8-羟基-2'-脱氧鸟苷(8-0HdG)常用于评价个体 DNA氧化损伤,是一个典型的生物标记物。利用溶胶凝胶法通过自组装技术分别将 Cu2+和 Zn2+固载到硅胶中,应用于复杂样品中痕量 8-OHdG的固相微萃取(SPME)在优化后的萃取条件下,结合高效液相色谱-紫外检测(HPLC-UV),构建了尿样中 8-OHdG的定量分析方法。结果表明,Cu2+和 Zn2+固载硅胶整体柱对 8-OHdG的富集倍数分别达到 61.64和51.20,具有良好的萃取性能;新建立SPME-HPLC-UV 分析检测方法的线性范围分别为0.91~100和1.22~100 nmol·L-1,R2>0.99,方法的检测限LODs(S/N=3)为 0.31 和 0.42 nmol·L-1,3个浓度加标实验显示,基于 Cu2+固载硅胶整体柱建立的分析方法加标回收率在 91.5%和 102.0%之间,日内和目间 RSD,在 2.7%至 9.7%之间(n=3),说明构建的 SPME-HPL.C-UV 分析方法可以灵敏、可靠、准确地检测实际尿样中的8-OHdG。
8-hydroxy-2'-deoxyguanosine (8-0HdG), as an typical biomarker, is commonly used to evaluate DNA damage, Cu2t and Zn2+ were successfully grafted to silica using solgel method through selfassembly technique, The extraction properties of the prepared monoliths were investigated when they were applied in solid-phase micro-extraction(SPME) of trace 8-OHdG in complex samples, Under the optimized extraction conditions, the analytical method of 8-0HdG in urine was estab.lished coupling with high performance liquid chromatography-ultraviolet( HPLC-UV). As the results showed, the extraction factors of the Cu2 and Zn2+ grafted silica monoliths for 8-OHdG were 61.64 and 51.20, respectively, which indicated an excellent extraction property. The linear ranges of established SPME-HPLC-UV analytical methods were within the rang of 0.91~100 and 1.22~100 nmol . L-1 with R2>0.99, respectively. Satisfactory sensitivity were obtained with the LODs(S/N=3) of 0.31 and 0.42 nmol .L-1. The spiking recoveries of 3 spiking level adding to real samples were from 91.5% to 102.0% based on the Cu2+ grafed silica proposed analysis method, The intra-day and inter-day RSDs were in the range of 2.7% and 9.7%(n=3). Successful applications illustrated the developed SPME-HPlC-UV analysis were sensitive, reliable, and accurate for detection of 8-OHdG in urinary sample.
